研究目的：

选择2020年12月至2021年12月龙华医院肛肠科门诊、病房及便秘门诊功能型便秘伴结肠黑变病者作为目标人群，分为2组，收集2组患者治疗后28天的症状评分及各项观察指标，并随访3个月，通过自身前后对照、组间对照，探究益气开秘方联合渗透性泻剂治疗功能性便秘伴结肠黑变病的临床有效性。通过收集功能型便秘伴和不伴结肠黑变病患者的基本信息、既往史、相关用药史等内容，分析功能性便秘患者发生结肠黑变病的危险因素；并招募10名正常人群，通过对比正常人群及益气开秘方联合渗透性泻剂治疗前后的功能性便秘伴结肠黑变病、功能性便秘不伴结肠黑变病的肠道菌群种类及代谢、肛门直肠压力和感觉阈值的不同，探究结肠黑变病可能的病理机制。

Objectives of Study：

Patients with functional constipation accompanied by melanosis colon were selected from the anorectal outpatient department, ward and constipation outpatient department of Longhua Hospital from December 2020 to December 2021 as the target population. They were divided into 2 groups. Symptom scores and observation indexes of patients in the 2 groups were collected 28 days after treatment, and followed up for 3 months. To explore the clinical effectiveness of yiqi decoction combined with osmotic purgative in the treatment of functional constipation with melanosis of colon. The risk factors for colon melanosis in functional constipation patients with and without functional constipation were analyzed by collecting basic information, past history and related medication history. And recruit 10 normal crowd, by comparing the normal crowd and yiqi open secret recipe combined osmotic laxatives before and after treatment of functional constipation with colon melanosis and functional constipation are not associated with the colon melanosis types and intestinal flora metabolism, the anorectal pressure and feel the different threshold, to explore the pathological mechanism of colon melanosis possible.

药物成份或治疗方案详述：

Description for medicine or protocol of treatment in detail：

纳入标准：

（1）符合《Rome Ⅵ-Functional GI Disorders: Disorders of Gut-Brain Interaction》功能型便秘； （2）肠镜明确诊断为结肠黑变病； （3）年龄18-75岁，男女不限； （4）符合气阴两虚中医证候诊断标准； （5）患者知晓了解本研究情况并自愿同意参加。

Inclusion criteria

(1) Functional constipation in accordance with Rome ⅵ -Functional GI Disorders: Disorders of Gut-Brain Interaction; (2) Colonoscopy confirmed the diagnosis of melanosis coli; (3) 18-75 years old, male or female; (4) Meet the criteria of TCM syndrome waiting diagnosis of qi and Yin deficiency; (5) Patients are aware of this study and voluntarily agree to participate.

排除标准：

（1）合并心血管、肝肾、造血系统等其他严重疾病者； （2）孕妇、妊娠期、哺乳期患者； （3）不能正确表达自己主诉者，如精神病、神经官能症、不能合作者； （4）治疗前3个月内曾参与其他药物研究。

Exclusion criteria：

(1) complicated with cardiovascular, liver, kidney, hematopoietic and other serious diseases; (2) pregnant women, pregnant women and lactation patients; (3) Those who cannot express their chief complaints correctly, such as mental illness, neurosis or inability to cooperate; (4) Participated in other drug studies within 3 months before treatment.
