探索针刺对遗忘型轻度认知障碍的临床疗效和中枢影像机制

Objectives of Study：

To explore the clinical effect and central imaging mechanism of acupuncture on amnestic mild cognitive impairment

药物成份或治疗方案详述：

Description for medicine or protocol of treatment in detail：

纳入标准：

符合 aMCI 诊断标准：（Perterson 2004 标准）： （1）记忆减退为主诉，且有知情者证实； （2）客观存在的记忆力下降且与年龄不符； （3）一般认知功能基本正常； （4）日常生活能力基本正常； （5）认知损害程度不足以诊断为痴呆。 ②有记忆障碍症状且病程>3 个月； ③简易精神状态检查量表(MMSE)评分大于等于 24 分； ④临床痴呆评定量表(CDR)总体的评分为 0.5，而记忆领域至少为 0.5； ⑤年龄 50-75 岁, 小学及以上文化程度； ⑥日常生活能力未受影响，量表 (ADL)评分≤26； ⑦右利手，汉族； ⑧愿意全程配合并签署知情同意书。

Inclusion criteria

Meet the aMCI diagnostic criteria: (Perterson 2004 criteria) : (1) Memory loss is the main complaint and it is confirmed by informed persons; (2) Objectively existing memory decline that is inconsistent with age; (3) General cognitive function is basically normal; (4) The ability of daily living is basically normal; (5) The degree of cognitive impairment is not sufficient to diagnose dementia. ② There are symptoms of memory impairment and the course of the disease is >3 months; ③ The score of the Simple mental State Examination Scale (MMSE) is greater than or equal to 24 points; The overall score of the Clinical Dementia Rating Scale (CDR) was 0.5 and the memory domain was at least 0.5; ⑤ Age 50-75 years old primary school education or above; The ability of daily living was not affected and the scale (ADL) score was ≤26; ⑦ Right-handed Han nationality; ⑧ Willing to cooperate and sign the informed consent.

排除标准：

①近 1 个月内曾接受认知功能的相关治疗者； ②近 1 月使用可能影响认知功能的药物，包括镇静剂，抗焦虑药，催眠药，促智 药和拟胆碱药； ③既往有脑卒中、脑肿瘤、帕金森病、脑炎、癫痫和脑外伤等疾病史； ④明显的白质病变（Fazekas 评分= 3–6）； ⑤严重精神疾病患者，嗜酒吸毒或其他精神性药物滥用者； ⑥严重听力、视力障碍和失语导致不能实现评估和不能完成试验者； ⑦有严重头部外伤史者，或其他累及中枢神经系统疾病，影响神经心理学量表评 估者； ⑧有严重心、肝、肾功能不全等全身性疾病者； ⑨有磁共振检查禁忌或者研究者认为其他不适合入组者

Exclusion criteria：

① Patients who have received cognitive function related treatment within the past 1 month; Use of drugs that may affect cognitive function in the past 1 month including sedatives anti-anxiety drugs hypnotics and psychotropic drugs Drugs and cholinomimetic drugs; (3) Previous history of stroke brain tumor Parkinson's disease encephalitis epilepsy and brain trauma; (4) Obvious white matter lesions (Fazekas score = 3-6); ⑤ Patients with serious mental illness alcoholics drugs or other psychotropic drug abusers; ⑥ Severe hearing visual impairment and aphasia resulting in inability to achieve the assessment and failure to complete the test; A history of severe head trauma or other diseases involving the central nervous system affect the neuropsychological scale assessment Estimator; ⑧ have serious heart liver renal insufficiency and other systemic diseases; ⑨ Patients with contraindications of MRI or other subjects deemed unsuitable by the researchers

本研究采用单盲法。研究过程中，除针灸医师外，患者、评估医师及统计人员等均不知分组情况。为确保盲法的成功实施，我们在研究开始前将对研究人员进行严格的培训，确保各个环节的分离原则。

Blinding：

Single blind method was used in this study. During the study except for acupuncturists patients evaluators and statisticians did not know the group status. In order to ensure the successful implementation of the blind method we will conduct strict training for researchers before the start of the study to ensure the separation principle.
