研究目的：

1通过非靶向血清代谢组学及宏基因组研究方法，明确抑郁症（轻中度）心脾两虚证、肝郁化火型及健康人群代谢物、肠道菌群和代谢通路特征。 2整合代谢组和宏基因组进行关联分析，探讨代谢物和肠道菌群在抑郁症（轻中度）心脾两虚证、肝郁化火型中可能的关联机制。

Objectives of Study：

1. By using non-targeted serum metabolomics and metagenomics research methods the metabolites gut microbiota and metabolic pathways of depression (mild to moderate) with heart-spleen deficiency syndrome liver fire syndrome and healthy individuals are clarified. 2. The association analysis between metabolites and gut microbiota is conducted by integrating metabolomics and metagenomics to explore the possible correlation mechanisms between metabolites and gut microbiota in depression (mild to moderate) with heart-spleen deficiency syndrome and liver fire syndrome.

药物成份或治疗方案详述：

Description for medicine or protocol of treatment in detail：

纳入标准：

1.符合DSM-V抑郁症的诊断标准； 2.符合中医辨证为心脾两虚证或肝郁化火证； 3. 年龄18－65岁，性别不限； 4.汉密尔顿抑郁量表（HAMD）17项评分为≥8分且≤24分，同时HAMD中抑郁情绪一项≥2分； 5.受试者同意并签署知情同意书。 同时符合上述5项者，方可纳入研究。

Inclusion criteria

1.Subjects must meet the diagnostic criteria for depression according to the DSM-V; only those who also meet the aforementioned five criteria will be included in the study. 2.Subjects should be diagnosed with Traditional Chinese Medicine as having either a deficiency of both the heart and spleen or a condition of liver qi stagnation transforming into fire. 3.Subjects must be aged between 18 and 65 years regardless of gender. 4.The Hamilton Depression Rating Scale (HAMD) score must be between ≥8 and ≤24 with a score of ≥2 on the depressive mood item of the HAMD. 5.Subjects must agree to and sign an informed consent form.

排除标准：

1.有自杀倾向，HAMD自杀项≥2分； 2.合并严重焦虑症状者，汉密尔顿焦虑量表（HAMA）≥29分； 3.有严重或不稳定的心、肝、肾、内分泌、血液等内科疾患者； 4.继发于躯体疾病的抑郁发作； 5.曾明确诊断为器质性精神障碍、精神分裂症、双相情感障碍； 6.精神活性物质（如酒精、烟草、阿片类等）和非成瘾物质所致的抑郁障碍； 7.一个月内服用过镇静催眠或抗抑郁等精神疾病药物（如利培酮、奥氮平等）、中成药（如逍遥丸、乌灵胶囊、疏肝解郁胶囊等）； 8.采样前3个月以内服用抗生素药物或益生元或胃肠动力药者； 9.采样前3个月内有腹泻、胃炎、消化道溃疡、炎症性肠炎等胃肠道疾病者； 10.3个月内接受过其它治疗抑郁症的临床试验者； 11.孕期及哺乳期妇女。 符合上述任意一项者，即予排除。

Exclusion criteria：

1.Subjects with suicidal tendencies indicated by a score of ≥2 on the HAMD suicide item. 2.Subjects with severe anxiety symptoms indicated by a Hamilton Anxiety Rating Scale (HAMA) score of ≥29. 3.Subjects with severe or unstable medical conditions affecting the heart liver kidneys endocrine system blood etc. 4.Depression episodes secondary to somatic diseases. 5.Subjects previously diagnosed with organic mental disorders schizophrenia or bipolar affective disorder. 6.Depression disorders induced by psychoactive substances (such as alcohol tobacco opioids etc.) and non-addictive substances. 7.Subjects who have taken sedative-hypnotic or antidepressant medications (such as risperidone olanzapine etc.) or traditional Chinese medicines (such as Xiao Yao Wan Wu Ling Capsule Shu Gan Jie Yu Capsule etc.) within the past month. 8.Subjects who have taken antibiotics probiotics or gastrointestinal motility medications within three months prior to sampling. 9.Subjects with gastrointestinal diseases such as diarrhea gastritis gastrointestinal ulcers or inflammatory bowel disease within three months prior to sampling. 10.Subjects who have participated in other clinical trials for depression treatment within the past three months. 11.Pregnant or breastfeeding women. Any subjects meeting any of the above criteria will be excluded from the study.

随机方法（请说明由何人用什么方法产生随机序列）：

本研究采用Excel软件中的随机函数生成随机数字，将随机数字依出现的先后，按 1、2、3……60排序。按抑郁症患者就诊的先后顺序进行排序，序号中对应的随机数字能被3整除不入组，其他的分配到病例观察组。同样的方法产生20例健康志愿者入组。

Randomization Procedure (please state who generates the random number sequence and by what method)：

In this study the random numbers were generated by the random function in Excel software and then sorted in ascending order according to their appearance i.e. 1 2 3 ... 60. The subjects with depression were sorted in the order of their visits to the clinic and the corresponding random numbers that could not be divided by 3 were excluded from the study group. The rest were allocated to the case observation group. Similarly 20 healthy volunteers were allocated to the study group using the same method.
