研究目的:

本研究拟观察上肢穴位电针和假电针对健康人的脑皮层影响,并利用EEG和fNIRS探究在不同频段功率谱,不同脑区/通道功能连接等方面的差异,证实电针的有效性,为后续临床卒中患者的上肢电针康复提供理论基础。

Objectives of Study:

This study investigates the cortical neuromodulatory effects of upper-limb acupoint electroacupuncture versus sham electroacupuncture in healthy adults through simultaneous EEG-fNIRS multimodal neuroimaging. By analyzing frequency-specific power spectral density alterations inter-regional functional connectivity patterns and neurovascular coupling dynamics we aim to elucidate the neurophysiological specificity of authentic electroacupuncture interventions. The findings will provide mechanistic evidence supporting the efficacy of electroacupuncture thereby establishing a neuroimaging-informed theoretical framework for optimizing upper-limb rehabilitation protocols in post-stroke patients.

Description for medicine or protocol of treatment in detail:

纳入标准:

(1)性别不限,18周岁≤年龄≤40周岁; (2)爱丁堡利手调查表(Edinburgh Handedness Inventory, EHI)测试为右利手; (3)受试者或其法定监护人(必要时)理解,自愿加入本研究试验,并签署《知情同意书》。

Inclusion criteria

Inclusion Criteria: No gender restrictions; age between 18 and 40 years (inclusive). Right-handedness confirmed by the Edinburgh Handedness Inventory (EHI). Participant or legal guardian (if applicable) fully understands and voluntarily agrees to participate in the study by signing the ​Informed Consent Form.

排除标准:

(1)神经系统疾病如癫痫、脑卒中、帕金森病、阿尔茨海默病等; (2)精神疾病如抑郁症、焦虑症、精神分裂症等; (3)1个月内服用任何精神科药物; (4)头颅不完整,曾行开颅手术和/或颅骨修补术; (5)严重的原发或继发性疾病包括心血管疾病)、糖尿病、肝肾功能不全等; (6)晕针晕血或有电针禁忌症,包括佩戴起搏器或嵌入式神经刺激器、心律失常、癫痫、孕妇或哺乳期妇女; (7)导电膏过敏; (8)正在进行其他临床试验或干预。

Exclusion criteria:

Exclusion Criteria: History of neurological disorders (e.g. epilepsy stroke Parkinsons disease Alzheimers disease or other major neurological pathologies). Diagnosed psychiatric conditions (e.g. depression anxiety disorders schizophrenia or other DSM-5/ICD-11 classified mental illnesses). Use of psychoactive medications (e.g. antidepressants anxiolytics antipsychotics) within 1 month prior to enrollment. Structural cranial abnormalities including skull defects history of craniotomy and/or cranial reconstruction surgery. Severe systemic diseases (e.g. uncontrolled cardiovascular disorders diabetes mellitus hepatic or renal insufficiency). Contraindications to electroacupuncture: Needle phobia hemophobia or vasovagal syncope tendencies; Implanted medical devices (e.g. pacemakers neurostimulators); Cardiac arrhythmia epilepsy pregnancy or lactation. Hypersensitivity to EEG conductive gel or electrode materials. Concurrent participation in other clinical trials or therapeutic interventions.

采用随机化方法,独立统计学家将使用 SPSS(版本 29.0)生成随机数字表。其中前1-12例接受A组干预,后13-24例接受B组干预,隔天A组接受B组干预,B组接受A组干预。符合条件的受试者将以 1:1 的比例随机分配到A组或B组。

Randomization Procedure (please state who generates the random number sequence and by what method):

A randomized crossover design will be implemented. An independent statistician will generate a randomization schedule using SPSS (Version 29.0). The first 12 participants will receive Intervention A followed by the next 12 participants receiving Intervention B. On the subsequent day the two groups will crossover interventions (Group A→Intervention B; Group B→Intervention A). Eligible participants will be randomly allocated to Group A or B at a 1:1 ratio through this block randomization sequence.

This mechanistic study does not utilize case report forms (CRFs).
