研究目的：

①初步探索并揭示复杂性肛瘘发病与愈合的机制。 ②通过严格设计的RCT研究验证加减胡连闭管汤促进复杂性肛瘘术后愈合的临床疗效。 ③初步阐释加减胡连闭管汤促进复杂性肛瘘术后愈合的疗效机制。

Objectives of Study：

①Preliminarily explore and reveal the mechanism of the pathogenesis and healing of complex anal fistula. ②To verify the clinical efficacy of modified Hu-lian-bi-guan formula in promoting postoperative healing of complex anal fistula through a rigorously designed RCT study. ③Preliminarily elucidate the efficacy mechanism of modified Hu-lian-bi-guan formula in promoting postoperative healing of complex anal fistula.

药物成份或治疗方案详述：

Description for medicine or protocol of treatment in detail：

纳入标准：

1: 符合复杂性肛瘘诊断及分类标准。2: 年龄18-70岁。3: 自愿参与本临床研究，并签署知情同意书。

Inclusion criteria

1: Meets the complex anal fistula diagnostic and categorization criteria. 2: Aged 18-70 years. 3: Voluntarily participate in this clinical study and sign an informed consent form.

排除标准：

1: 合并严重心、脑、肾等重要脏器疾病。2: 存在手术禁忌症，不能接受或耐受手术。3: 对试验用药存在药物过敏。4: 精神情志异常患者。5: 既往曾行肛周手术者。6: 妊娠及哺乳期妇女或在研究期间不能避孕者。7: 研究者认为不适合参加该研究的其他情况。

Exclusion criteria：

1: Combination of serious heart brain kidney and other important organ diseases. 2: Presence of contraindications to surgery inability to accept or tolerate surgery. 3: Presence of drug allergy to the experimental drugs. 4: Patients with abnormal mental mood. 5: Those who have had perianal surgery in the past. 6: Pregnant and lactating women or those who are unable to use contraception during the study period. 7: Other conditions that the investigator considers unsuitable for participation in the study

数据采集和管理（说明：数据采集和管理由两部分组成，一为病例记录表(Case Record Form, CRF)，二为电子采集和管理系统(Electronic Data Capture, EDC)，如ResMan即为一种基于互联网的EDC：

（1）培训：课题开始前对所有课题研究人员进行培训，培训内容包括：课题实施步骤及方法，诊断、纳入、排除标准；CRF和各个量表的评价及填写方法； （2）数据质控：本课题的原始数据位病例报告表、知情同意书、临床病历和原始化验单，原始数据必须有可溯源性。研究者手工填写病例报告表，定期收集，并采用双人双录入方法进行数据录入。 （3）数据备份：原始数据在按要求完成数据录入和核查后，按编号的顺序归档保存，并填有检索目录等，以备查考。电子数据文件包括数据库、检查程序、分析程序、分析结果、编码本和说明文件等，应分类保存，并有多个备份保存于不同磁盘或记录介质上，妥善保存，防止损坏。

Data collection and Management (A standard data collection and management system include a CRF and an electronic data capture：

(1) Training: Before the start of the project all researchers involved in the project will undergo training which includes: the implementation steps and methods of the project diagnostic inclusion and exclusion criteria; the evaluation and completion methods of CRF and various scales. (2) Data Quality Control: The original data for this project includes case report forms informed consent documents clinical medical records and original laboratory reports. The original data must be traceable. Researchers manually complete the case report forms which are collected regularly and entered using a double data entry method. (3) Data Backup: After the original data is entered and verified according to requirements it is archived in numerical order and includes a retrieval directory for reference. Electronic data files including databases check programs analysis programs analysis results coding books and explanatory documents should be saved in categories with multiple backups stored on different disks or recording media properly preserved to prevent damage.
