研究目的：

1）探究针灸足三里（ST36）对老年人群的抗炎及延缓衰老的作用，为针灸抗炎、延缓衰老提供循证依据，为延缓衰老探寻新的方向和手段； 2）揭示针灸足三里(ST36)抗衰老作用的生物分子机制。

Objectives of Study：

1) To explore the anti-inflammation and anti-aging effect of acupuncture and moxibustion Zusanli (ST36) on the elderly provide evidence based evidence for anti inflammation and anti-aging of acupuncture and moxibustion and explore new directions and means for anti-aging; 2) To reveal the biological and molecular mechanism of anti-aging effect of acupuncture and moxibustion Zusanli (ST36).

Description for medicine or protocol of treatment in detail：

纳入标准：

（1）年龄在45~70岁的健康志愿者，性别不限； （2）患者认知功能正常，无沟通障碍，无严重基础性疾病及精神疾病； （3）自愿参加项目并签署知情同意书。

Inclusion criteria

(1) Healthy volunteers aged 45-70 regardless of gender; (2) The patient has normal cognitive function no communication barriers no serious underlying diseases or mental illnesses; (3) Volunteer to participate in the project and sign an informed consent form.

排除标准：

（1）既往有严重凝血功能障碍，合并严重心血管、肝、肾功能不全等脏器严重病变，或严重躯体疾病或精神障碍症状者； （2）近4周内接受过可能会影响本研究结果的药物或非药物治疗患者； （3）穴位周围局部皮肤感染、过敏或穴位周围疾病的患者； （4）既往曾出现针灸治疗的禁忌证，如晕针等； （5）已参加其他临床试验的志愿者。

Exclusion criteria：

(1) Previous severe coagulation dysfunction combined with severe cardiovascular liver kidney dysfunction and other organ diseases or severe physical or mental disorders symptoms; (2) Patients who have received medication or non medication treatment within the past 4 weeks that may affect the results of this study; (3) Patients with local skin infections allergies or diseases around acupoints; (4) In the past there have been contraindications to acupuncture and moxibustion treatment such as dizziness; (5) Volunteers who have participated in other clinical trials.

1. 干预方法： 参照十四五规范教材《经络腧穴学》中穴位定位方法取穴，取足阳明胃经双侧足三里（ST36），对足三里进行针灸干预，由针灸医生进行针灸操作，参照“十四五”规划教材《刺法灸法学》，受试者取仰卧或坐位，在针刺腧穴局部皮肤及针灸师手部用75%酒精常规消毒后，使用0.25 mm×50 mm一次性无菌针灸针（华佗，苏州医疗用品厂有限公司，中国），直刺穴位20-30mm后，针灸师进行提刺或旋转操作，以患者局部有酸麻胀感为得气，将18mm×200mm艾条（汉医，南阳汉医艾绒有限责任公司，中国）对双侧足三里穴位进行悬灸温和灸，艾灸时间为30min，至皮肤出现红晕为度。结束后用无菌医用干棉签按压针孔拔针。 2. 干预频率： 针灸足三里（ST36）单穴干预每周2次，持续8周，共16次。

研究采取独立样本设计试验方法，统计人员对基线和结局评价不知情，干预治疗师对于结局评价不知情。由统计人员采用统计软件成随机数字表，然后统一交给研究协调人员。研究人员将受试者分配到随机号码。

The study adopted an independent sample design experimental method with statisticians unaware of baseline and outcome evaluations and intervention therapists unaware of outcome evaluations. The statisticians use statistical software to create a random number table which is then uniformly handed over to the research coordinator. The researchers assigned the subjects to random numbers.

数据采集和管理（说明：数据采集和管理由两部分组成，一为病例记录表(Case Record Form, CRF)，二为电子采集和管理系统(Electronic Data Capture, EDC)，如ResMan即为一种基于互联网的EDC：

研究涉及的标本及信息资料的使用、检测，将在国内某非外资背景测序公司检测，后续将进行公开招标，未涉及外资背景研究机构/企业。研究结束后保留的剩余标本、影像资料及其他数据将研究对象同意后保留，可用于将来的研究。剩余样本将回收至衰老与肿瘤研究中心低温储藏室，在知情同意中说明这些标本与资料在研究结束后将可能用于其他相关研究。评估数据与标本的计算和使用将在符合研究计划、伦理要求内进行，审阅的文件（如CRF、调查问卷、视频或音频记录、标本跟踪日志）将安排专门的人员进行半个月检查一次，质控人员为本次研究的主要研究员。

Data collection and Management (A standard data collection and management system include a CRF and an electronic data capture：

The use and testing of the specimens and information materials involved in the study will be conducted by a non foreign background sequencing company in China. Subsequent public bidding will be conducted. If no foreign background research institution/enterprise is involved the remaining specimens imaging materials and other data retained after the study is completed will be retained with the consent of the research subjects for future research. The remaining samples will be collected to the low-temperature storage room of the Aging and Oncology Research Center and it will be stated in the informed consent that these samples and data may be used for other related studies after the end of the study. The calculation and use of evaluation data and specimens will be carried out within the scope of compliance with the research plan and ethical requirements. The reviewed documents (such as CRF survey questionnaire video or audio records specimen tracking logs) will be inspected by dedicated personnel every half month and quality control personnel will be the main researchers of this study.
