研究目的：

本研究以无先兆偏头痛患者为研究对象，观察针刺对无先兆偏头痛患者临床症状改善的同时，利用fMRI技术分别观察针刺组、药物组治疗前后对无先兆偏头痛患者大脑功能活动及功能连接网络的影响；利用肠道菌群指标分别分析针刺与氟桂利嗪治疗对无先兆偏头痛患者肠道菌群的物种注释、物种组成分析、样本比较分析、物种差异分析等，比较辨证针刺与氟桂利嗪对肠道菌群种类和丰度影响的共性与差异性，分别提取与无先兆偏头痛患者疗效改善密切相关的脑功能连接和肠道菌群生物学标记，分析患者大脑功能网络变化与肠道菌群调控的相关性，揭示针刺和氟桂利嗪治疗无先兆偏头痛的脑-肠-微生物机制，为临床研究提供指导。

Objectives of Study：

In this study, patients with migraine without aura was included to observe acupuncture on the improvement of clinical symptoms, and use fMRI technique to observe the brain function and functional connection network of patients migraine without aura before and after treatment in acupuncture group and drug group. Based on analyzing the effects of acupuncture and flunarizine on the intestinal flora, species composition, sample comparison, species difference of the intestinal flora of patients without migraine, comparing the commonality and difference of the effects of acupuncture and flunarizine groups on the species and abundance of intestinal flora and extracting brain function and intestinal flora biomarkers, which were closely related to the improvement of the efficacy of patients without migraine, to analyze correlation between changes in brain function network and regulation of intestinal flora and revealing the brain-intestinal-microbial mechanism of acupuncture and flunarizine treatment on migraine without aura for providing guidance of clinical study.

药物成份或治疗方案详述：

Description for medicine or protocol of treatment in detail：

纳入标准：

(1)年龄在18-55周岁,性别不限; (2)右利手，受教育年限≥6年; (3)有1年以上偏头痛病史，偏头痛首发在50岁之前; (4)在过去的3个月及基线，偏头痛发作≥2次，且每月发作天数<15天; (5)完成基线头痛日记;(6)本人签署知情同意书。

Inclusion criteria

(1) men or women aged 18–55 years; (2) right handed, length of education >=6 years; (3) a history of migraine of at least 1 year with the first onset before the age of 50 years; (4) migraine attacks >=2 times and number of migraine days <15 days per month during the past 3 months and baseline observation; (5) completion of the baseline headache diary; (6) provision of written informed consent.

排除标准：

(1)合并有头部外伤史、继发性头痛、紧张型头痛、间歇性头痛、丛集型头痛，以及心血管和消化系统疾病的患者; (2)既往经历过主要肠道切除手术或胃肠道大手术者; (3)有精神疾病、感染、出血疾病或过敏体质者; (4)孕妇、哺乳期妇女或计划在6个月内怀孕者; (5)不能理解或记录头痛日记; (6)存在核磁禁忌症，如幽闭恐怖症; (7)过去3个月内使用过抗生素或接受针刺治疗，并持续3天或以上; (8)长期使用止痛药物和预防偏头痛的药物; (9)过去3个月内参加类似的研究。

Exclusion criteria：

(1) any combination of head trauma history, secondary headache, tension-type headache, intermittent-type headache, cluster-type headache, or cardiovascular and digestive diseases; (2) a history of major intestinal resection or gastrointestinal surgery; (3) presence of psychiatric disease, infection, bleeding disorders, or allergic constitution; (4) pregnancy, lactation, or plans to become pregnant within 6 months; (5) inability to understand or fill in a headache diary; (6) nuclear magnetic contraindications, such as claustrophobia; (7) a history of taking antibiotics or receiving acupuncture therapy for 3 days or more in the past 3 months; (8) long-term use of analgesic drugs and medications to prevent migraine attacks; (9) participation in a similar study in the last 3 months.

数据采集和管理（说明：数据采集和管理由两部分组成，一为病例记录表(Case Record Form, CRF)，二为电子采集和管理系统(Electronic Data Capture, EDC)，如ResMan即为一种基于互联网的EDC：

CRF表由研究人员进行采集并管理。临床数据采用SAS或SPSS统计分析系统进行统计，影像数据采用SPM软件、Matlab软件平台等进行分析 ；肠道菌群数据采用PE300测序策略，在Illunima Miseq平台上进行高通量测序。

CRF forms are collected and managed by researchers.The clinical data were statistically analyzed by SAS or SPSS statistical analysis system; the image data was analyzed by SPM software and Matlab software platform；the PE300 sequencing strategy will be adopted for intestinal flora data and high-throughput sequencing will be conducted on the Illunima Miseq platform.
