研究目的：

本课题拟开展临床研究对高龄女性患者进行电针干预，观察GV期卵母细胞纺锤体及染色体分布、非整倍体分析，进一步以单细胞 RNA-seq技术为基础，验证各组卵母细胞SAC调控E3连接酶APC/CCDC20相关基因及蛋白表达差异；探讨电针治疗对高龄老化卵母细胞减数的影响，基于SAC对E3连接酶APC/CCDC20的调控机制阐明电针促进高龄老化卵母细胞减数分裂顺利发生、降低卵子非整倍体率的作用机制，为中医药在辅助生殖领域应用提供新的思路和治疗靶点。

Objectives of Study：

This project intends to carry out clinical research on electroacupuncture intervention in elderly female patients, observe the spindle and chromosome distribution of GV stage oocytes, and analyze aneuploidy, and further verify each group of oocytes based on single-cell RNA-seq technology SAC regulates E3 ligase APC/CCDC20-related gene and protein expression differences; explores the effect of electroacupuncture on the reduction of aging oocytes, and clarifies that electroacupuncture promotes aging oocytes based on the regulation mechanism of SAC on E3 ligase APC/CCDC20 The mechanism of the smooth occurrence of cell meiosis and the reduction of the rate of egg aneuploidy provides new ideas and therapeutic targets for the application of traditional Chinese medicine in the field of assisted reproduction.

药物成份或治疗方案详述：

Description for medicine or protocol of treatment in detail：

纳入标准：

① 符合不孕症诊断标准：夫妻双方有正常性生活，且未避孕1年未妊娠者; ② 拟在曙光医院接受IVF-ET治疗的患者，年龄在35-42岁之间； ③ 符合中医肾精亏虚型诊断标准； 肾精亏虚型不孕症诊断标准：(参照全国中医药行业高等教育“十二五”规 划教材《中医妇科学》及高等医药院校教材《中医诊断学》第5版) 主症：婚久不孕，月经先后不定期，月经量少色淡，甚或闭经。 次症：腰膝酸软，头晕耳鸣，性欲减退，神疲乏力，健忘脱发。 舌脉：舌淡红，苔白，脉沉细。 具备婚久不孕，主症具备1项，次症具备1-2项，参照舌脉即可诊断。 ④ 签署知情同意书。

Inclusion criteria

① Meet the diagnostic criteria for infertility: Both spouses have normal sexual life and have not been pregnant for 1 year without contraception; ② Patients who intend to receive IVF-ET treatment in Shuguang Hospital, aged between 35-42 years old; ③ Meet the diagnostic criteria of TCM kidney essence deficiency type; Diagnostic criteria for infertility due to deficiency of kidney essence: (Refer to the Twelfth Five-Year Plan Planning textbook "Chinese Medicine Gynecology" and higher medical college textbook "Chinese Medicine Diagnostics" 5th edition) Main symptoms: long-term marriage, infertility, irregular menstruation, low menstrual flow, pale color, or even amenorrhea. Secondary symptoms: weakness in the waist and knees, dizziness, tinnitus, decreased libido, fatigue, forgetfulness and hair loss. Tongue and pulse: pale red tongue, white coating, deep and thin pulse. With long-term marriage and infertility, the main symptom has 1 item, and the secondary symptom has 1-2 items, which can be diagnosed by referring to the tongue and pulse. ④ Sign the informed consent form.

排除标准：

① 不符合诊断标准者； ② 因服用激素、免疫抑制剂等药物、卵巢恶性肿瘤放化疗引起卵巢功能减退者; ③ 合并严重心血管、肝、肾和造血系统等严重原发疾病者。

Exclusion criteria：

① Those who do not meet the diagnostic criteria; ② Those who have ovarian dysfunction caused by taking hormones, immunosuppressive drugs, radiotherapy and chemotherapy for malignant ovarian tumors; ③ Patients with severe primary diseases such as severe cardiovascular, liver, kidney and hematopoietic system.
