研究目的：

（1）通过对比针刺组和假针刺组PMI患者的临床症状、情绪状态改善情况及（The Hypothalamic-Pituitary-Adrenal axis, HPA轴）相关激素及肠道菌群变化情况，探索针刺治疗PMI的临床效应。 （2）使用“大尺度脑功能网络分析中基于种子点的功能连接分析法”，对比针刺组和假针刺PMI患者下丘脑-全脑功能连接的变化，及脑功能连接变化与临床效应和HPA轴相关激素及肠道菌群的相关性分析，揭示针刺治疗PMI的中枢整合机制。

Objectives of Study：

(1) To explore the clinical effects of acupuncture in treating PMI by comparing the clinical symptoms improvement of emotional state and changes of hormones and intestinal flora related to the HPA axis in PMI patients in the acupuncture group and the sham-acupuncture group. (2) Using the seed-based functional connectivity analysis in large-scale brain functional network analysis we compared the changes in hypothalamic-whole-brain functional connectivity between the acupuncture group and the sham-acupuncture PMI patients and analyzed the correlation between the changes in brain functional connectivity and the clinical effects and the changes in hormones of the HPA axis and and intestinal flora to reveal the central integration mechanism of the acupuncture treatment of PMI. central integration mechanism.

药物成份或治疗方案详述：

Description for medicine or protocol of treatment in detail：

纳入标准：

①女性，45岁≤年龄≤60岁，接受教育≥6年，右利手；②符合STRAW+10的诊断标准，月经紊乱，或最后一次月经（Final Menstrual Period，FMP）大于>1年；③ 符合 ICSD-3 中慢性失眠的诊断标准；④匹兹堡睡眠质量指数（Pittsburgh Sleep Quality Index，PSQI）评分≤7分；⑤进入研究前至少两周内未服用任何药物；⑥自愿参加本次研究并签署知情同意书。

Inclusion criteria

① Female 45 ≤ age ≤ 60 ≥ 6 years of education right-handed; ② meeting the diagnostic criteria of STRAW+10 menstrual disorders or Final Menstrual Period (FMP) > > 1 year; ③meeting the diagnostic criteria of ICSD-3 ④ Pittsburgh Sleep Quality Index (PSQI) score ≤ 7; ⑤ not taking medication for at least two weeks prior to entering the study and ⑥ voluntarily participating in this study and signing an informed consent form.

排除标准：

1)有高血压、糖尿病、冠心病等系统性疾病病史者；2)合并有心脑血管、肝、肾和造血系统等严重危及生命的原发性疾病者；3)意识不清，不能表达主观不适症状者； 4)酗酒和（或）精神药物滥用和依赖（含安眠药）所致失眠者；5)伴有重度抑郁、焦虑症状者； 6)伴有严重头痛、偏头痛，头部外伤史及手术史者；7)抗凝剂使用史或镇静剂或抗焦虑药的常规使用史；8)既往有磁共振扫描禁忌者，如：幽闭恐惧症、金属植入、纹身等。

Exclusion criteria：

1) those with a history of systemic diseases such as hypertension diabetes mellitus coronary artery disease etc.; 2) those with comorbidities of cardiovascular cerebrovascular hepatic renal and hematopoietic systems and other serious and life-threatening primary diseases; 3) those who are unconscious and are unable to express the subjective symptoms of discomfort; 4) those who have insomnia due to alcoholism and/or psychotropic substance abuse and dependence (including sleeping pills); 5) those who are accompanied by symptoms of severe depression and anxiety; and 6) those who are accompanied by severe headaches migraine history of head trauma and history of surgery; 7) history of anticoagulant use or routine use of sedatives or anxiolytics; 8) previous contraindications to magnetic resonance scanning e.g. claustrophobia metal implants tattoos etc.

1个独立的研究者使用PASS 15.0 将48例病人随机分为针刺组和假针刺组，随机结果保存在密封不透明的信封中，最后由治疗者打开信封，根据随机结果进行干预。

Eligible patients will be randomly allocated in a 1:1 ratio to the acupuncture or sham acupuncture group. An independent researcher who will not be involved in contact of patients generates the random sequence by PASS 15.0 (NCSS LLC. Kaysville Utah USA ncss.com/software/pass.).
