研究目的：

在临床研究基础上，观察真针刺、假针刺对慢性失眠伴情绪障碍患者脑网络功能连接改变，为科学阐释针刺治疗慢性失眠伴情绪障碍的作用机制提供新思路，以期作为针刺通过调节脑网络相关脑区功能，治疗慢性失眠伴情绪障碍的前瞻性研究。

Objectives of Study：

On the basis of clinical research this study observes the changes in brain network functional connectivity induced by true acupuncture and sham acupuncture in patients with chronic insomnia accompanied by emotional disturbances. The goal is to provide new insights for scientifically elucidating the mechanisms underlying acupuncture's therapeutic effects on chronic insomnia with emotional disorders. This research aims to serve as a pioneering study demonstrating how acupuncture may regulate the functions of brain regions involved in brain networks to effectively treat chronic insomnia accompanied by emotional disturbances.

纳入标准：

1.慢性失眠伴情绪障碍患者纳入标准： ①符合上述的诊断标准； ②年龄18岁至60岁者，小学及以上学历，为右利手者； ③近1个月内未参加其它临床研究； ④无磁共振扫描禁忌（安装心脏起博器及神经刺激器、体内金属异物、幽闭恐惧症等）； ⑤理解试验过程，签署患者知情同意书，自愿选择参加试验的受试者。 注：同时符合上述5项标准的患者，方可纳入研究。 2.健康受试者纳入标准： ①年龄18岁至60岁者，小学及以上学历，为右利手者； ②经相关体检，各项指标均在正常范围，既往未确诊功能性或器质性疾病；无精神、神经系统的家族遗传病史； ③无磁共振扫描禁忌（安装心脏起博器及神经刺激器、体内金属异物、幽闭恐惧症等）； ④理解试验过程，签署患者知情同意书，自愿选择参加试验的受试者。 注：同时符合以上4项标准的健康受试者，方可纳入。

Inclusion criteria

1. Inclusion Criteria for Patients with Chronic Insomnia Accompanied by Emotional Disturbances: ① Meet the diagnostic criteria mentioned above; ② Ages 18 to 60 with at least a primary school education and right-handed; ③ Have not participated in other clinical studies within the past month; ④ Have no contraindications for magnetic resonance scanning (e.g. pacemaker or neurostimulator implants metallic foreign bodies in the body claustrophobia etc.); ⑤ Understand the experimental procedure sign the informed consent form and voluntarily choose to participate as a subject. Note: Only patients meeting all 5 criteria above may be included in the study. 2. Inclusion Criteria for Healthy Subjects: ① Ages 18 to 60 with at least a primary school education and right-handed; ② Passed relevant physical examinations with all indicators within the normal range; no prior diagnosis of functional or organic diseases; no family history of psychiatric or neurological genetic diseases; ③ Have no contraindications for magnetic resonance scanning (e.g. pacemaker or neurostimulator implants metallic foreign bodies in the body claustrophobia etc.); ④ Understand the experimental procedure sign the informed consent form and voluntarily choose to participate as a subject. Note: Only healthy subjects meeting all 4 criteria above may be included.

排除标准：

1.慢性失眠伴情绪障碍患者排除标准： ①伴有肝肾功不全、肿瘤或循环、消化、内分泌、呼吸等系统严重基础疾病者；癫痫发作史或家族史、 脑器质性疾病及严重躯体疾病史者；既往己诊为重度抑郁症、双向情感障碍、精神分裂症或其他精神障碍类疾病者； ②伴有慢性头痛、偏头痛、头部外伤史及头部占位性病变者； ③妊娠期、哺乳期妇女； ④酗酒或药物滥用者；凝血功能障碍或严重疤痕体质者。 注：凡符合上述4项标准中任何一项的患者，则予以排除。 2.健康受试者排除标准： ①妊娠期、哺乳期妇女； ②既往头部外伤史者； ③近1个月内参加其它临床试验者。 注：凡符合上述任何一项的健康受试者，即予以排除。

Exclusion criteria：

1. Exclusion Criteria for Patients with Chronic Insomnia Accompanied by Emotional Disturbances: ① Patients with liver or kidney dysfunction tumors or severe underlying diseases affecting the circulatory digestive endocrine or respiratory systems; a history or family history of epilepsy; cerebral organic diseases; severe physical illnesses; or a previous diagnosis of major depression bipolar disorder schizophrenia or other psychiatric disorders; ② Patients with chronic headaches migraines a history of head trauma or intracranial space-occupying lesions; ③ Pregnant or breastfeeding women; ④ Alcoholics or individuals with drug abuse issues; patients with coagulation disorders or severe scarring tendencies. Note: Any patient meeting one of the 4 criteria above will be excluded from the study. 2. Exclusion Criteria for Healthy Subjects: ① Pregnant or breastfeeding women; ② Individuals with a history of head trauma; ③ Subjects who have participated in other clinical trials within the past month. Note: Any healthy subject meeting one of the criteria above will be excluded from the study.

随机方法（请说明由何人用什么方法产生随机序列）：

由独立的统计人员应用SAS软件编程产生随机号码。将纳入的慢性失眠伴情绪障碍受试者按照1:1比例分为两组。具体方法为：将写有随机数字、组别的信件装入不透光的牛皮信封中密封，信封上标注序号，当纳入慢性失眠伴情绪障碍患者时，按入组顺序打开序号相同的信封，随机分配到各组进行试验。随机人员负责保存随机信封，不参与分组及后期试验数据采集的具体实施。

Randomization Procedure (please state who generates the random number sequence and by what method)：

The random numbers were generated by independent statisticians using SAS software programming. The enrolled subjects with chronic insomnia accompanied by emotional disorders were divided into two groups in a 1:1 ratio. The specific method involved: writing the random numbers and group assignments on letters which were then sealed in opaque envelopes labeled with corresponding serial numbers. When patients with chronic insomnia accompanied by emotional disorders were enrolled the envelopes labeled with the same serial numbers as the enrollment order were opened in sequence and the patients were randomly assigned to one of the two groups for the trial. The personnel responsible for randomization were in charge of storing the randomization envelopes and did not participate in the group allocation or the specific implementation of subsequent trial data collection.
