利用EEG技术，筛选出taVNS治疗DOC的脑电特征指标。

Objectives of Study：

Using EEG technology to screen for EEG characteristic indicators for taVNS treatment of DOC

药物成份或治疗方案详述：

我们计划纳入慢性意识障碍患者50例参与本项研究。每名患者将接受为期4周的taVNS治疗，治疗前后我们将采集每名患者的脑电图和CRS-R评分。4周taVNS治疗后，我们将会在治疗后的第一个月、第三个月、第六个月和第十二个月对患者进行随访。我们根据CRS-R评分是否增加将50例患者分为应答组和无应答组。比较两组入组前的脑电图在脑电微状态、功率谱和功能连接的差异，筛选出taVNS治疗DOC的脑电特征指标。

Description for medicine or protocol of treatment in detail：

We plan to include 50 patients with chronic consciousness disorders in this study. Each patient will undergo a 4-week taVNS treatment and we will collect EEG data and CRS-R scores before and after the treatment. After the 4-week taVNS treatment we will follow up with the patients at the first month third month sixth month and twelfth month. Based on whether the CRS-R score increases the 50 patients will be divided into an responder group and a non-responder group. We will compare the differences in EEG microstates power spectra and functional connectivity before treatment between the two groups and identify EEG characteristic indicators for taVNS treatment of DOC.

纳入标准：

（1）符合慢性意识障碍诊断标准；（2）年龄18-70岁，性别不限；（3）患病1个月以上，常规治疗方法促醒失败；（4）心、肺、肝、肾功能基本正常；（5）评估适宜（神经影像、脑电图、各种诱发电位）；（6）患者存在失匹配负波；（7）预期生存时间大于3个月；（8）能合作观察不良事件和疗效；（9）患者其法定代理人签署书面知情同意书；（10）女性患者妊娠试验是阴性；（11）凝血功能正常。

Inclusion criteria

(1) Meets the diagnostic criteria for disorders of consciousness ; (2) Aged 18-70 years no gender restriction; (3) Disease duration of more than 1 month with failure of conventional awakening treatments; (4) Basic normal heart lung liver and kidney function; (5) Suitable for assessment (neuroimaging EEG various evoked potentials); (6) Presence of mismatch negativity (MMN) in EEG; (7) Expected survival time greater than 3 months; (8) Able to cooperate in monitoring adverse events and treatment efficacy; (9) The patient or their legal guardian signs a written informed consent form; (10) Female patients have a negative pregnancy test; (11) Normal coagulation function.

排除标准：

（1）神经退行性疾病、颅内感染及脑肿瘤术后昏迷患者；（2）入组前1月内意识持续改善或恶化者；（3）伴严重并发症者；（4）试验前三个月内参加过其他临床试验的患者；（5）严重过敏史或过敏体质的患者；（6）有其他严重的内科疾患或严重不可控制的感染；（7）吸毒、药物滥用、长期嗜酒以及艾滋病患者。

Exclusion criteria：

(1) Patients with comas due to neurodegenerative diseases intracranial infections or post-brain tumor surgery; (2) Patients whose consciousness has shown sustained improvement or deterioration within 1 month prior to enrollment; (3) Patients with severe complications; (4) Patients who have participated in other clinical trials within the past three months; (5) Patients with a history of severe allergies or allergic constitution; (6) Patients with other severe medical conditions or serious uncontrolled infections; (7) Patients with substance abuse drug addiction chronic alcoholism or HIV.
