研究目的：

（1）通过随机对照研究，聚焦伴有胰岛素抵抗的PCOS患者，与二甲双胍对照，评价加味葛根芩连汤改善PCOS患者胰岛素抵抗促进排卵的有效性；并对不良反应和不良事件进行分析，以突出安全性的优势，为中医药治疗PCOS提供临床循证证据。 （2）通过随机对照研究，评价比较加味葛根芩连汤配方颗粒与传统饮片，在改善PCOS患者胰岛素抵抗促进排卵疗效和安全的一致性，为经典方剂配方颗粒与饮片汤剂的比较提供有力证据。

Objectives of Study：

(1) A randomized controlled study focused on PCOS patients with insulin resistance and compared with metformin to evaluate the effectiveness of modified Gegenqinlian Decoction in improving insulin resistance and promoting ovulation in PCOS patients; The adverse reactions and adverse events were analyzed to highlight the advantages of safety and provide clinical evidence-based evidence for the treatment of PCOS by traditional Chinese medicine. (2) A randomized controlled study was conducted to evaluate and compare the consistency of the efficacy and safety of modified Gegen Qinlian Decoction formula granules and traditional decoction pieces in improving insulin resistance and promoting ovulation in PCOS patients providing strong evidence for the comparison between classical prescription formula granules and decoction pieces.

药物成份或治疗方案详述：

（1）基础治疗：生活方式干预 （2）干预分组 ①中药配方颗治疗组： 在生活方式干预的基础上，服用加味葛根芩连汤（葛根、黄芩、黄连等）配方颗粒。由一方制药提供，每次一包，用100ml温水溶解冲服，每天一次，共治疗12周。 ② 中药传统饮片治疗组：在生活方式干预的基础上，服用加味葛根芩连汤中药饮片。饮片全部采用标准化煎煮技术，每天2次，每次1包，共服用12周。 ③ 二甲双胍对照组：在生活方式干预的基础上，加用二甲双胍缓释片，根据患者胰岛素抵抗的情况，每次0.5-1g ，每日1次，餐中服用。共服用12周。两组均在月经第1-3天开始服药。

Description for medicine or protocol of treatment in detail：

(1)Basic Treatment: Lifestyle Intervention (2) Intervention Grouping ① Treatment Group of Traditional Chinese Medicine Formula Granules: On the basis of lifestyle intervention patients should take the formula granules of Modified Gegen Qinlian Decoction (Gegen Scutellaria baicalensis Huanglian etal). Provided by Yifang Pharmaceutical one pack each time dissolved and flushed with 100ml warm water once a day for a total of 12 weeks. ② Treatment Group of Traditional Chinese Medicine Decoction Pieces : On the basis of lifestyle intervention patients in this group should take the Modified Gegen Qinlian Decoction traditional Chinese medicine decoction pieces. The decoction pieces all adopted standardized decoction technology 2 times a day 1 pack each time. ③ Metformin control group: On the basis of lifestyle intervention patients took metformin sustained-release tablets. According to the patient's insulin resistance 0.5 - 1 g each time once a day with meals. Take a total of 12 weeks. Both groups started to take the drug on the first 1 - 3 days of menstruation.

1: 同时符合PCOS和胰岛素抵抗诊断标准 2: 年龄18-39岁 3: 自愿签署知情同意书者

Inclusion criteria

排除标准：

1: 先天性肾上腺皮质增生或存在分泌雄激素的肿瘤病史者 2: 库欣综合征者 3: 高泌乳素血症以及甲状腺功能异常等 4: 体重指数(BMI)≥35kg/m² 5: 吸烟患者 6: 最近1个月内使用过口服避孕药（如达英-35）及胰岛素增敏剂（如二甲双呱、毗格列酮）或注射胰高血糖素样肽-1受体激动剂如利拉鲁肽、杜拉糖肽等 7: 具有心血管疾病、高血压、糖尿病、肝脏、肾脏等重要脏器慢性疾病患者 8: 妊娠期、哺乳期及治疗期间有生育要求的患者

Exclusion criteria：

1: People with Congenital adrenal hyperplasia or history of androgen secreting tumors 2: People with Cushing's syndrome 3: People with hyperprolactinemia and thyroid dysfunction 4: Body mass index (BMI)≥35kg/m² 5: Smokers 6: In the last 1 month she has used oral contraceptives (such as danyin-35) and insulin sensitizers (such as dimethyldiquide pilaglitazone) or injected glucagon-like peptide-1 receptor agonists (such as liraglutide dura glycotide etc.) 7: Patients with cardiovascular disease hypertension diabetes liver kidney and other chronic diseases of important organs 8: Patients with fertility requirements during pregnancy lactation and treatment

随机化分配方法由广东省中医院临床方法学团队研究人员完成，采用简单随机方法，随机方案先依次编写好的数字顺序表（此顺序即为以后进入临床试验的合格受试者的序号），通过SAS9.4统计软件的PROCPLAN过程编程产生随机数字。
