研究目的：

本研究拟以芒针透刺慢性前列腺炎患者（CP）为研究对象，采用弥散张量成像和静息态功能磁共振结合的多模态成像技术，以下丘脑神经环路为切入点，采集针刺前后CP患者在芒针治疗前后的影像数据，同时引入先进的脑连接组学图论分析技术，通过多模态影像数据与效应指标、体液生化指标的多维度相关分析，探讨芒针透刺疗法对CP患者下丘脑神经网络的调控机制。以期更深入揭示针刺治疗CP的作用机制，对促进针灸治疗CP临床推广有着重要的科学意义和实用价值。

Objectives of Study：

This study aims to investigate the mechanism of elongated needle therapy on the hypothalamic neural network in chronic prostatitis (CP) patients by using a multi-modal imaging technique combining diffusion tensor imaging and resting-state functional magnetic resonance imaging with the hypothalamic neural circuit as the entry point. The imaging data of CP patients before and after mung bean acupuncture treatment were collected and advanced brain connectomics graph theory analysis techniques were introduced. Through multi-dimensional correlation analysis of multi-modal imaging data effect indicators and biochemical indicators the study aims to explore the regulatory mechanism of elongated needle therapy on the hypothalamic neural network in CP patients. The study aims to provide a deeper understanding of the mechanism of acupuncture treatment for CP which has important scientific significance and practical value for promoting the clinical application of acupuncture for CP.

Description for medicine or protocol of treatment in detail：

纳入标准：

（1）符合中医精浊诊断标准； （2）符合西医慢性非细菌性前列腺炎的诊断标准； （3）年龄 20～50 岁； （4）病程≥3 个月； （5）NIH-CPSI 前列腺炎症状评分总积分大于 4； （6）自愿加入本临床研究，并签署知情同意书。 注：同时符合上述 6 项的受试者，方可纳入本项研究。

Inclusion criteria

(1) Those who do not meet the inclusion criteria; (2) Patients under the age of 20 or over the age of 50; (3) suffering from serious cardiovascular cerebrovascular liver kidney diseases; (4) Patients who are unconscious suffer from mental disorders or are accompanied by mental disorders; (5) Patients who have taken painkillers or sedatives within 1 week before treatment; (6) Patients with contraindications to MRI scanning; (7) Are participating in other clinical investigators. Note: Patients meeting any of the above criteria were excluded.

排除标准：

（1）不符合纳入标准者； （2）年龄在 20 岁以下或 50 岁以上的患者； （3）患有严重的心脑血管、肝、肾疾病者； （4）意识不清，患有精神疾患，或伴有心理障碍的患者； （5）治疗前 1 周内服用过止痛药、镇静药者； （6）有 MRI 扫描禁忌症者； （7）正在参加其他临床研究者。 注：凡符合上述任何一条的患者，均予以排除。

由第三方将60名慢性前列腺炎受试者按照随机数字表，随机分为 2 组，每组 30 例。将序号、随机数字、组别装入不透光的牛皮信封中密封，信封上标注序号，当纳入慢性前列腺炎受试者时，按入组顺序打开序号相同的信封，随机分配到各组进行试验。

Randomization Procedure (please state who generates the random number sequence and by what method)：

60 subjects with chronic prostatitis were randomly divided into two groups by a third party according to the random number table with 30 cases in each group. The serial number random number and group were sealed in opaque cowhine envelopes and the serial number was marked on the envelope. When subjects with chronic prostatitis were enrolled the envelopes with the same serial number were opened in the order of enrollment and randomly assigned to each group for the trial.

数据采集和管理（说明：数据采集和管理由两部分组成，一为病例记录表(Case Record Form, CRF)，二为电子采集和管理系统(Electronic Data Capture, EDC)，如ResMan即为一种基于互联网的EDC：

本研究按照临床研究数据管理SOP进行数据录入与管理，并采用CRF表与电子Excel的CRF版本相结合的方式上传、保存数据。患者试验周期结束后1周内进行数据核对，确认CRF填写正确，且无缺项、漏项。如需进行更改，须按照要求进行注明或修改。数据由独立的评估人员进行双份录入并校对，针对不同的录入结果，应逐项核对原始的纸质CRF表，保证数据录入的准确性。录入工作结束后，随机抽取10份电子版CRF与纸质版CRF逐项对比，确保数据录入与原始版本一致。确认无误后，将所有的数据封存。

Data collection and Management (A standard data collection and management system include a CRF and an electronic data capture：

In this study data entry and management were carried out according to the SOP of clinical research data management and the CRF form was combined with the CRF version of electronic Excel to upload and save the data. The data of patients were checked within 1 week after the end of the trial period to confirm that the CRF was filled in correctly and there were no missing items and missing items. If any changes are required they must be noted or modified as required. Data were entered in duplicate and proofread by an independent assessor and the original paper CRF form should be checked item by item for different entry results to ensure the accuracy of data entry. After the entry work 10 electronic CRFS were randomly selected to compare with the paper CRFS item by item to ensure that the data entry was consistent with the original version. After confirmation all data will be sealed.
